Abstract: To devise immunodiagnostics of phytoviruses, monoclonal antibodies (Mab's) of the IgG and IgM classes to Arabis mosaic virus (AMV) have been prepared and conjugated with horse-raddish peroxidase. The subclass of Mab's and their antigen binding constants have been determined. To find an optimal way of the virus diagnostics in purified preparation and plant juice, variants of the direct and indirect ELISA have been compared. The sensitivity of the ELISA diagnostics has been 2–3 ng AMV per milliliter. We have also developed one-step latex test with very similar sensitivity, applicable to AMV determination.
